Disruption of XIST expression in mammary epithelial cells leads to chromatin changes on the inactive X chromosome, transcriptional reactivation of X-linked genes, and defective luminal differentiation. Genomic DNA was isolated from HMLE cells and sequenced to identify heterozygous SNPs on the X chromosome in order to evaluate the transcriptional and epigenetic status of the X chromosome upon XIST deletion.
