Here, we reported a strategy of obtaining functionally competent induced HSCs (iHSCs) from human pluripotent stem cell by mimicking the embryonic development of HSCs in vivo. RNAseq results showed that the iHSCs share similar gene expression pattern with quiescent primary HSCs (qHSCs) such as hSTAP, desmin, GFAP, PDGFRb, etc.Our induction strategy provided a novel and robust method to efficiently generate functional iHSCs from human pluripotent stem cell line differentiation, which could be used as a model for hepatocyte toxicity prediction, anti-liver fibrosis drug screening and hepatitis B and C virus infection induced liver fibrosis.
