Homo sapiens loci: Modification of genomic ATM on-target and off-target sites by evolved TALENs. Homo sapiens strain:HEK293 or U2OS-GFP

To evaluate the DNA cleavage specificity of canonical TALENs and evolved TALENs targeting the ATM gene in the human genome, we sequenced on-target and off-target sites of ATM genomic target sites in human tissue culture treated with TALENs. The target site and its corresponding known off-target sites were amplified from genomic DNA isolated from either HEK293 or U2OS-GFP cells transfected with plasmids expressing various TALENs targeting the ATM site.
