To define the cell types expressing Esr1 in the developing mouse cortex, we applied scRNA-seq from P1 neocortex expressing the transgenic reporter. Droplet-based scRNA-seq was performed with both GFP+ (cellular lineages expressing Esr1) and unsorted, whole populations. Esr1-Cre;LSL-Sun1-GFP pups at P1 were used for this experiment. The entire neocortex was dissected - cells were isolated by Fluorescence-activated cell sorting (FACS) with only DAPI as the unsorted population; or isolated by FACS with DAPI and GFP to enrich for the Esr1+ population only. Cells collected from each condition were processed and analyzed using scRNA-seq
