A disturbance of histone protein composition renders nuclear DNA immunogenic, inducing a type I interferon response to self. Overall design: We investigated histone pre-mRNA misprocessing in patient and control fibroblasts using RNA-seq. Total RNA was extracted from the cells and transcripts remaining after an initial selection for polyadenylated RNA were ribo-depleted, and both poly(A)+ and poly(A)- fractions then sequenced.
