Single guide RNA CRISPR /Cas(sg) editing 3 nucleotides and prime editing (pe) single nucleotide of CArG element in Tspan2 promoter effect on transcriptome in aortas

Aortas from sgCArG WT versus sgCArG homozygous mice at 12 wks and peCArG WT and peCArG homozygenous mutant mice at 8 weeks were isolated for total RNA extraction. RNA samples were then subjected to deep sequencing to profile gene expression in aortas. We will compare the transcriptome of aortas from sgCArG WT versus sgCArG mutant as well as peCArG WT versus peCArG mutant to determine the genes consistently regulated by CArG mutation of TSPAN2 gene in mice.本研究分离了12周龄sgCArG野生型（sgCArG WT）与sgCArG纯合突变小鼠的主动脉，以及8周龄peCArG野生型（peCArG WT）与peCArG纯合突变小鼠的主动脉，用于总RNA提取。随后对获取的RNA样本进行深度测序以解析主动脉的基因表达谱。本研究将对比sgCArG野生型与sgCArG突变型、peCArG野生型与peCArG突变型小鼠主动脉的转录组，以此确定小鼠中受TSPAN2基因CArG突变持续调控的基因。
