Hepatic transcriptome profiling of liver-specific Xbp1 knockout mice fed a high fat diet

Xbp1 is an important regulator of unfolded protein response and lipid metabolism. Its dyregulation has been associcated in human NASH. Feeding a high fat diet with fructose/sucrose to mice causes progressive, fibrosing steatohepatitis. This study is to use RNA-Seq to identify differentially expressed genes in hepatic Xbp1 deficient mice livers fed with a high fat diet compared to controls. Hepatic Xbp1 deficient mice or flox controls were fed either regular chow or a high fat diet (n=4). Samples from each cohort were pooled into two replicates.
X盒结合蛋白1(Xbp1)是未折叠蛋白反应与脂质代谢的关键调控因子。其异常调控与人类非酒精性脂肪性肝炎(NASH)密切相关。给小鼠饲喂添加果糖/蔗糖的高脂饮食,可诱发进展性纤维化脂肪性肝炎。本研究采用RNA测序(RNA-Seq)技术,对比高脂饮食喂养的肝脏Xbp1缺陷小鼠与对照小鼠的肝脏组织,以鉴定差异表达基因。实验中,肝脏Xbp1缺陷小鼠及其flox对照小鼠分别喂食普通饲料或高脂饮食(每组n=4)。每一组队列的样本均被合并为两个重复样本。
