Genome-wide Profiling of Adenine Base Editor Specificity by EndoV-seq. Homo sapiens

EndoV-seq utilizes Endonuclease V (EndoV) to nick the inosine (I)-containing DNA strand of genomic DNA that has been deaminated by ABE in vitro. The treated DNA is then whole-genome sequenced to identify overlapping ends generated by EndoV and Cas9 nickase (nCas9). Genomic sites are scored based on DNA reads with overlapping ends to identify both on-target sites and off-target sites.
